3-Fluorobenzoic hydrazide (CAS number 499-55-8) is a hydrazide building block, derived from benzoic acid. Hydrazide derivatives are bioactive, for example isoniazid which is an antibiotic for tuberculosis. The hydrazide moiety in 3-fluorobenzoic hydrazide is nucleophilic and readily reacts with electrophiles, such as isothiocyanate derivatives. The resulting hydrazide-isothiocyanate products can undergo further cyclization, forming triazole-thione derivatives. The triazole-thione derivatives synthesized in this manner show antimicrobial activity, with a minimum inhibitory concentration (MIC) of 62.5 µg/mL. Additionally, they have low toxicity against human cells. The heterocycles synthesized from 3-fluorobenzoic hydrazide allow for late-stage functionalization, owing to their multiple functionalities.
